Scope
This part of ISO 12966 provides an overview of the gas chromatographic determination of free and bound fatty acids in animal and vegetable fats and oils, which follows their conversion to fatty acid methyl esters (FAME). The qualitative and quantitative determination of the composition of fatty acids using gas-liquid chromatography (GLC) is a frequently used method in lipid analysis. It is used to characterize fats and oils or fatty foods after extraction of the oil from the matrix. The bound fatty acids of the triacylglycerols (TAG) and, depending on the esterification process, the free fatty acids (FFA) and other lipids are converted into fatty acid methyl esters (FAME). can be determined using capillary gas chromatography. Depending on the number of different fatty acids (theoretically there can be more than 50 different fatty acids), capillary columns with a length of 10 m to 100 m are used for separation. The GLC from FAME is suitable for natural and synthetic mixtures of tri-, di- and monoacylglycerols, for fatty acid esters, free fatty acids, soaps and other fatty compounds. With this series of standards, FAME from C4 to C26 including saturated fatty acid methyl ester, monounsaturated cis and trans fatty acid methyl ester and polyunsaturated cis and trans fatty acid methyl ester can be determined. Isopropyl and butyl esters are often used to determine short-chain fatty acids in order to avoid interference with the solvent peak and to reduce differences in detector responses.
