This technical standard is applicable to the detection of serum neutralizing antibodies of West Nile virus-susceptible animals. Through the detection of serum West Nile virus neutralizing antibodies, it can assist in determining the status of animals infected with West Nile virus or the immune effect of animal West Nile fever vaccines. The main technical contents are: 1. Construction of a cell line stably expressing West Nile virus C-prM-E protein; 2. Construction of a West Nile virus replicon that lacks cellular protein genes and expresses green fluorescent protein; 3. West Nile virus Packaging and titration of viral replicon viruses; 4. Collection and inactivation of animal serum; 5. Neutralization of serum and West Nile virus replicon virus and cell inoculation; 6. Observation and determination of results through fluorescence focus; 7. Pass Observation and determination of viral plaque counting results.
