Scope
The purified Tremella polysaccharide was prepared, and the total sugar content was determined by the phenol sulfuric acid method. Free monosaccharides, oligosaccharides, and hexoses, pentoses, and uronic acids (or toluene derivatives) in polysaccharides can be dehydrated under the action of concentrated sulfuric acid to form furfural or hydroxymethylfurfural, which can be condensed with phenol to form an orange-yellow color. Compound, within a certain range, its color depth (absorbance in the visible light region) is proportional to the concentration of sugar. Hexose has a maximum absorption peak at a wavelength of 490nm (pentose and uronic acid at 480nm), so colorimetry can be used to Measure at the corresponding wavelength, and calculate the total sugar content in the sample based on the standard curve of standard monosaccharides.
