Scope
This standard specifies a method for estimating soil microbial biomass by measuring the total amount of extractable organic matter after killing fresh soil microorganisms by fumigation with chloroform. This standard only describes the determination of extractable organic carbon content, but this method is also applicable to the estimation of microbial nitrogen content and microbial ninhydrin reaction nitrogen content in soil. This standard applies to the determination of microbial biomass in aerobic and anaerobic (waterlogged, paddy fields) soils with a wide range of soil pH. This standard is also applicable to the determination of microbial biomass in supersaturated soil containing easily decomposable organic substrates and potassium sulfate solution. NOTE: Chloroform fumigation also affects soil fauna, but the carbon contribution of such organisms is generally small (<5%) and usually negligible.
